NTRK1 (HGNC:8031): (neurotrophic receptor tyrosine kinase 1) This gene encodes a member of the neurotrophic tyrosine kinase receptor (NTKR) family. This kinase is a membrane-bound receptor that, upon neurotrophin binding, phosphorylates itself and members of the MAPK pathway. The presence of this kinase leads to cell differentiation and may play a role in specifying sensory neuron subtypes. Mutations in this gene have been associated with congenital insensitivity to pain, anhidrosis, self-mutilating behavior, cognitive disability and cancer. Alternate transcriptional splice variants of this gene have been found, but only three have been characterized to date. [provided by RefSeq, Jul 2008]

The missense c.2141G>A(p.Gly714Asp) variant in NTRK1 gene has been reported in compound heterozygous state in an individual affected with Congenital insensitivity to pain with anhidrosis (Wang Q, et. al., 2015). This variant has been reported to the ClinVar database as Uncertain significance. The p.Gly714Asp variant is novel (not in any individuals) in gnomAD Exomes and 1000 Genomes database. The amino acid change p.Gly714Asp in NTRK1 is predicted as conserved by GERP++ and PhyloP across 100 vertebrates. The amino acid Gly at position 714 is changed to a Asp changing protein sequence and it might alter its composition and physico-chemical properties. Since the variant has been reported in only a single patient it has been classified as Uncertain Significance.
