The encoded nuclear protein may also bind to and enhance the DNA joining activity of HIV-1 integrase. This gene has been found to be a tumor suppressor, and mutations in it have been associated with malignant rhabdoid tumors. Alternatively spliced transcript variants have been found for this gene. This variant has been observed in individual(s) with schwannomas and/or meningiomas (PMID: 20930055, 22038540). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 30202). This variant is not present in population databases (ExAC no frequency). This sequence change replaces proline with leucine at codon 48 of the SMARCB1 protein (p.Pro48Leu). The proline residue is highly conserved and there is a moderate physicochemical difference between proline and leucine.
