Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.8e-05 in 1209456 control chromosomes, including 6 hemizygotes. The observed variant frequency is approximately 1.65 fold of the estimated maximal expected allele frequency for a pathogenic variant in DMD causing Dystrophinopathies phenotype (1.1e-05). To our knowledge, no occurrence of c.8159G>A in individuals affected with Dystrophinopathies and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 409916). Based on the evidence outlined above, the variant was classified as likely benign. -
