ClinVar contains an entry for this variant (Variation ID: 480442). This missense change has been observed in individual(s) with breast cancer (PMID: 25452441). -
The p.I82N variant (also known as c.245T>A), located in coding exon 3 of the RAD50 gene, results from a T to A substitution at nucleotide position 245. The isoleucine at codon 82 is replaced by asparagine, an amino acid with dissimilar properties. This alteration has been detected in 1/1824 patients with triple negative breast cancer who were unselected for a family history of breast or ovarian cancer (Couch FJ et al. J. Clin. Oncol., 2015 Feb;33:304-11). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
