Variant summary: CACNA2D2 c.782C>T (p.Pro261Leu) results in a non-conservative amino acid change located in the VWA N-terminal domain (IPR013608) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 250148 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.782C>T has been reported in the literature in a compound heterozygous individual affected with Cerebellar Atrophy With Seizures And Variable Developmental Delay (Butler_2018). These data do not allow clear conclusions about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication have been ascertained in the context of this evaluation (PMID: 30410802). ClinVar contains an entry for this variant (Variation ID: 530501).
