The p.T1349A variant (also known as c.4045A>G), located in coding exon 9 of the BRCA1 gene, results from an A to G substitution at nucleotide position 4045. The threonine at codon 1349 is replaced by alanine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -

In summary, this variant is a novel missense change that is not predicted to affect protein function.
