NM_017617.5:c.2753A>G
Variant summary
Our verdict is Likely benign. The variant received -3 ACMG points: 2P and 5B. PP3_ModerateBP6BS2
The NM_017617.5(NOTCH1):c.2753A>G(p.Asn918Ser) variant causes a missense change. The variant allele was found at a frequency of 0.0000366 in 1,612,970 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a pathogenic outcome for this variant. Variant has been reported in ClinVar as Conflicting classifications of pathogenicity (no stars). Synonymous variant affecting the same amino acid position (i.e. N918N) has been classified as Likely benign.

Adams-Oliver syndrome 5 Uncertain:1Benign:1
Familial thoracic aortic aneurysm and aortic dissection Uncertain:1
The p.N918S variant (also known as c.2753A>G), located in coding exon 18 of the NOTCH1 gene, results from an A to G substitution at nucleotide position 2753. The asparagine at codon 918 is replaced by serine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
Aortic valve disease 1 Uncertain:1
