The c.1510G>A (p.E504K) alteration is located in exon 14 (coding exon 13) of the PLEKHG5 gene. This alteration results from a G to A substitution at nucleotide position 1510, causing the glutamic acid (E) at amino acid position 504 to be replaced by a lysine (K). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 504 of the PLEKHG5 protein (p.Glu504Lys). This variant is present in population databases (rs552364487, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with PLEKHG5-related conditions.
