Variant summary: The BRIP1 c.2477A>G (p.Asn826Ser) variant located in the P-loop containing nuceloside triphosphate hydrolase domain (via InterPro) causes a missense change involving a conserved nucleotide with 3/4 in silico tools (SNPs&GO not captured here due to low reliability index) predict a damaging outcome, although these predictions have yet to be functionally assessed. The variant of interest was observed in the large, broad control population, ExAC, with an allele frequency of 1/119650, which does not exceed the estimated maximal expected allele frequency for a pathogenic BRIP1 variant of 1/16000. The variant of interest has not, to our knowledge, been reported in affected individuals via publications and/or reputable databases/clinical diagnostic laboratories. Therefore, until additional information becomes available, the variant of interest has been classified as a "Variant of Uncertain Significance (VUS)." -
The frequency of this variant in the general population, 0.000008 (2/249882 chromosomes, http://gnomad.broadinstitute.org), is uninformative in assessment of its pathogenicity. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with ovarian cancer (PMID: 33008098). This variant has been identified in 2/249882 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
The p.N826S variant (also known as c.2477A>G), located in coding exon 16 of the BRIP1 gene, results from an A to G substitution at nucleotide position 2477. The asparagine at codon 826 is replaced by serine, an amino acid with highly similar properties. This variant was identified in multiple cohorts with features consistent with a hereditary breast and/or ovarian cancer syndrome (Barbosa A et al. Cancers (Basel), 2020 Sep;12; van der Merwe NC et al. Front Oncol, 2022 Dec;12:938561; Faria JP et al. Breast Cancer Res Treat, 2024 Oct;207:615-624). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
