The encoded protein is thought to be a calcium-dependent cell-adhesion protein that is primarily expressed in the brain. Mutations in this gene on human chromosome X are associated with sporadic infantile epileptic encephalopathy and to a female-restricted form of epilepsy (EFMR; also known as PCDH19RE). It was not observed in approximately 6,400 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Although the c.1521delC variant has not been previously reported to our knowledge, many other frameshift variants downstream of this position have been reported in the Human Gene Mutation Database in association with PCDH19-related disorders (Stenson et al., 2014). We interpret the c.1521delC deletion as a pathogenic variant. - |
