The c.1271C>T (p.S424L) alteration is located in exon 9 (coding exon 7) of the IRF6 gene. This alteration results from a C to T substitution at nucleotide position 1271, causing the serine (S) at amino acid position 424 to be replaced by a leucine (L). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This variant was reported in individuals with features consistent with IRF6-related disorders; in at least one individual, it was determined to be de novo (Desmyter, 2010; Matsuzawa, 2010). This amino acid position is highly conserved in available vertebrate species. This alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, this alteration is classified as likely pathogenic. -
