The p.K2716I variant (also known as c.8147A>T), located in coding exon 54 of the RYR2 gene, results from an A to T substitution at nucleotide position 8147. The lysine at codon 2716 is replaced by isoleucine, an amino acid with dissimilar properties. This variant was detected in a cardiomyopathy/arrhythmia genetic testing cohort; however, clinical details were limited, and additional cardiac variants were detected in some cases (van Lint FHM et al. Neth Heart J, 2019 Jun;27:304-309). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
