The p.V723L variant (also known as c.2167G>T), located in coding exon 13 of the ATM gene, results from a G to T substitution at nucleotide position 2167. The valine at codon 723 is replaced by leucine, an amino acid with highly similar properties. This variant was detected in a cohort of 1663 Brazilian breast cancer patients who underwent hereditary multigene panel testing (Guindalini RSC et al. Sci Rep, 2022 Mar;12:4190). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
This missense variant replaces valine with leucine at codon 723 of the ATM protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373).
