This sequence change replaces arginine, which is basic and polar, with leucine, which is neutral and non-polar, at codon 712 of the POLE protein (p.Arg712Leu). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with POLE-related conditions. ClinVar contains an entry for this variant (Variation ID: 581851). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt POLE protein function. The p.R712L variant (also known as c.2135G>T), located in coding exon 19 of the POLE gene, results from a G to T substitution at nucleotide position 2135. The arginine at codon 712 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
