The serine at codon 2414 is replaced by leucine, an amino acid with dissimilar properties. A homology-directed repair activity assay showed that this alteration has no effect on BRCA2 function (Guidugli L et al. Cancer Res. 2013 Jan 1;73(1):265-75). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
