The encoded protein shows slow kinetics of activation and inactivation, and is necessary for the cardiac pacemaking process. This channel may also mediate responses to sour stimuli. Mutations in this gene have been linked to sick sinus syndrome 2, also known as atrial fibrillation with bradyarrhythmia or familial sinus bradycardia. This missense change has been observed in individual(s) with clinical features of HCN4-related conditions (PMID: 25145517, 25145518, 26206080, 26688388, 28104484, 30471092). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 374859). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt HCN4 protein function. Experimental studies have shown that this missense change affects HCN4 function (PMID: 25145517, 25145518). For these reasons, this variant has been classified as Pathogenic. -
