Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 246170 control chromosomes. c.74G>A has been reported in the literature as a homozygous genotype in at-least one individual affected with Chronic Granulomatous Disease (example, Koker_2013 cited in Roos_2021, Akar_2021). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 33629196, 23910690, 36241643, 34547651). No other clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014.
