The protein represents a conventional non-muscle myosin; it should not be confused with the unconventional myosin-14 (MYO14). Myosins are actin-dependent motor proteins with diverse functions including regulation of cytokinesis, cell motility, and cell polarity. Mutations in this gene result in one form of autosomal dominant hearing impairment. Multiple transcript variants encoding different isoforms have been found for this gene. The Pro552Leu variant in MYH14 gene has not been previously reported in individu als with hearing loss and was absent from large population studies. Computationa l prediction tools and conservation analysis suggest that this variant may impac t the protein; though this information is not predictive enough to determine pat hogenicity. The variant is located in the last three bases of the exon, which is part of the 5? splice region. Computational tools do not suggest an impact to s plicing; however, this data is not predictive enough to rule out pathogenicity. In summary, the clinical significance of the Pro552Leu variant is uncertain. -
