Genes affected
GLI2 (HGNC:4318): (GLI family zinc finger 2) This gene encodes a protein which belongs to the C2H2-type zinc finger protein subclass of the Gli family. Members of this subclass are characterized as transcription factors which bind DNA through zinc finger motifs. These motifs contain conserved H-C links. Gli family zinc finger proteins are mediators of Sonic hedgehog (Shh) signaling and they are implicated as potent oncogenes in the embryonal carcinoma cell. The protein encoded by this gene localizes to the cytoplasm and activates patched Drosophila homolog (PTCH) gene expression. It is also thought to play a role during embryogenesis. The encoded protein is associated with several phenotypes- Greig cephalopolysyndactyly syndrome, Pallister-Hall syndrome, preaxial polydactyly type IV, postaxial polydactyly types A1 and B. Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Aug 04, 2024 | The c.1541A>C (p.Y514S) alteration is located in exon 10 (coding exon 10) of the GLI2 gene. This alteration results from a A to C substitution at nucleotide position 1541, causing the tyrosine (Y) at amino acid position 514 to be replaced by a serine (S). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. - |
Holoprosencephaly 9;C4014479:Postaxial polydactyly-anterior pituitary anomalies-facial dysmorphism syndrome Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | May 31, 2017 | In summary, this variant has uncertain impact on GLI2 function. The available evidence is currently insufficient to determine its role in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). Family studies have indicated that an individual, with clinical features consistent with GLI2-related disease, inherited this variant from an unaffected parent. This suggests that the variant is not likely a primary cause of disease. This variant is not present in population databases (ExAC no frequency). This sequence change replaces tyrosine with serine at codon 514 of the GLI2 protein (p.Tyr514Ser). The tyrosine residue is highly conserved and there is a large physicochemical difference between tyrosine and serine. - |
