This sequence change replaces arginine with leucine at codon 15 of the SCN3A protein (p.Arg15Leu). The arginine residue is highly conserved and there is a moderate physicochemical difference between arginine and leucine. This variant is present in population databases (rs139769668, ExAC 0.006%). This variant has not been reported in the literature in individuals affected with SCN3A-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
