This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 295 of the AGBL5 protein (p.Asp295Asn). This variant is present in population databases (no rsID available, gnomAD 0.003%). This missense change has been observed in individuals with retinitis pigmentosa (PMID: 26720455; internal data). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 242932). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. This variant disrupts the p.Asp295His amino acid residue in AGBL5.
