The valine at codon 215 is replaced by leucine, an amino acid with highly similar properties. Although this specific alteration has not been reported in the literature, another alteration (c.643G>C) that results in the same amino acid change at position 215 (p.V215L) was identified in an cohort of 1058 unselected individuals with colon cancer (Yurgelun MB et al. J. Clin. Oncol., 2017 Apr;35:1086-1095). This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
