The c.1403G>C (p.R468P) alteration is located in exon 4 (coding exon 4) of the MSH6 gene. This alteration results from a G to C substitution at nucleotide position 1403, causing the arginine (R) at amino acid position 468 to be replaced by a proline (P). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
This missense variant replaces arginine with proline at codon 468 of the MSH6 protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function (internally defined REVEL score threshold >= 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with endometrial cancer (PMID: 27443514). This variant has been identified in 2/282540 chromosomes in the general population by the Genome Aggregation Database (gnomAD).
