Variant summary
Our verdict is Uncertain significance. The variant received 2 ACMG points: 2P and 0B. PM2
The NM_005732.4(RAD50):c.2108C>G(p.Ser703Cys) variant causes a missense change. The variant allele was found at a frequency of 0.00000658 in 152,054 control chromosomes in the GnomAD database, with no homozygous occurrence. Variant has been reported in ClinVar as Uncertain significance (★★). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. S703Y) has been classified as Uncertain significance.

Hereditary cancer-predisposing syndrome Uncertain:1
The p.S703C variant (also known as c.2108C>G), located in coding exon 13 of the RAD50 gene, results from a C to G substitution at nucleotide position 2108. The serine at codon 703 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
