The c.3781A>G (p.I1261V) alteration is located in exon 32 (coding exon 31) of the WRN gene. This alteration results from a A to G substitution at nucleotide position 3781, causing the isoleucine (I) at amino acid position 1261 to be replaced by a valine (V). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Werner syndrome Uncertain:1
This sequence change replaces isoleucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 1261 of the WRN protein (p.Ile1261Val). This variant is present in population databases (rs201661079, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with WRN-related conditions. ClinVar contains an entry for this variant (Variation ID: 238162). An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign".
