The encoded protein is a member of the MRE11/RAD50 double-strand break repair complex which consists of 5 proteins. The histidine at codon 45 is replaced by tyrosine, an amino acid with similar properties. This variant was observed in 1/7051 unselected female breast cancer patients and was not observed in 11241 female controls of Japanese ancestry (Momozawa Y et al. Nat Commun, 2018 Oct;9:4083). This variant was also detected in a cohort of 236 female breast cancer patients from India (Mittal A et al. Ecancermedicalscience, 2022 Aug;16:1434). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
