Mutations in this gene are associated with X-linked Alport syndrome, also known as hereditary nephritis. Like the other members of the type IV collagen gene family, this gene is organized in a head-to-head conformation with another type IV collagen gene so that each gene pair shares a common promoter. This sequence change replaces serine, which is neutral and polar, with phenylalanine, which is neutral and non-polar, at codon 1488 of the COL4A5 protein (p.Ser1488Phe). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with Alport syndrome (PMID: 8940267). This variant is also known as 4665C>T.
