AGL (HGNC:321): (amylo-alpha-1, 6-glucosidase, 4-alpha-glucanotransferase) This gene encodes the glycogen debrancher enzyme which is involved in glycogen degradation. This enzyme has two independent catalytic activities which occur at different sites on the protein: a 4-alpha-glucotransferase activity and a amylo-1,6-glucosidase activity. Mutations in this gene are associated with glycogen storage disease although a wide range of enzymatic and clinical variability occurs which may be due to tissue-specific alternative splicing. The variant allele was found at a frequency of 8e-06 in 250684 control chromosomes (gnomAD). c.276delG has been reported in the literature in individuals affected with Glycogen Storage Disease Type III (Lucchiari_2002, Crushell_2010). These data indicate that the variant may be associated with disease. At least one publication reports this variant results in <10% of normal AGL activity (Crushell_2010). One ClinVar submitter (evaluation after 2014) cites the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. - |
