The new acceptor site was used in 97% of the transcripts resulting in a frameshift and premature protein truncation (PMID: 34846068). This variant has been reported in individuals affected with breast cancer (PMID: 33471991; Color internal data). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Loss of PALB2 function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Likely Pathogenic. -
