Variant summary: CFTR c.325_327delinsG (p.Tyr109GlyfsX4) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. The variant was absent in 251124 control chromosomes. c.325_327delinsG has been reported in the literature in multiple individuals affected with Cystic Fibrosis, including as a compound heterozygous genotype (e.g. Terzic_2019). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 31523618). ClinVar contains an entry for this variant (Variation ID: 53696). Based on the evidence outlined above, the variant was classified as pathogenic.
