Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt SCN1A protein function. ClinVar contains an entry for this variant (Variation ID: 961713). This variant is also known as c.Tyr779Phe. This missense change has been observed in individual(s) with clinical features of generalized epilepsy with febrile seizures plus (GEFS+) as well as focal epilepsy with auditory features (PMID: 17679682, 28202706, 30977726). In at least one individual the variant was observed to be de novo. This variant is present in population databases (rs121918782, gnomAD 0.004%). This sequence change replaces tyrosine, which is neutral and polar, with phenylalanine, which is neutral and non-polar, at codon 790 of the SCN1A protein (p.Tyr790Phe). -
