This missense variant replaces glycine with arginine at codon 288 of the RAD51D protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. In a case-control study, this variant was detected in 1/3429 invasive early-onset ovarian cancer cases and absent in 2,772 controls (PMID: 26261251). This variant has been identified in 2/251440 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
The p.G288R variant (also known as c.862G>C), located in coding exon 9 of the RAD51D gene, results from a G to C substitution at nucleotide position 862. The glycine at codon 288 is replaced by arginine, an amino acid with dissimilar properties. In one case-control study, this variant was identified in 1/3429 individuals with epithelial ovarian cancer and in 0/2772 unaffected controls (Song H et al. J. Clin. Oncol. 2015 Sep;33:2901-7). This variant was also detected in a cohort of 1663 Brazilian breast cancer patients who underwent hereditary multigene panel testing (Guindalini RSC et al. Sci Rep, 2022 Mar;12:4190). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -

This sequence change replaces glycine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 288 of the RAD51D protein (p.Gly288Arg). This variant is present in population databases (no rsID available, gnomAD 0.003%). This missense change has been observed in individual(s) with breast cancer and/or ovarian cancer (PMID: 26261251, 35264596). ClinVar contains an entry for this variant (Variation ID: 472625). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt RAD51D protein function with a negative predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
