This sequence change inserts 1 nucleotide in exon 1 of the MSH2 mRNA (c.153dupG), causing a frameshift at codon 52. This creates a premature translational stop signal (p.Leu52Alafs*30) and is expected to result in an absent or disrupted protein product. -

The c.153dupG pathogenic mutation, located in coding exon 1 of the MSH2 gene, results from a duplication of G at nucleotide position 153, causing a translational frameshift with a predicted alternate stop codon (p.L52Afs*30). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
