BARD1 (HGNC:952): (BRCA1 associated RING domain 1) This gene encodes a protein which interacts with the N-terminal region of BRCA1. In addition to its ability to bind BRCA1 in vivo and in vitro, it shares homology with the 2 most conserved regions of BRCA1: the N-terminal RING motif and the C-terminal BRCT domain. The RING motif is a cysteine-rich sequence found in a variety of proteins that regulate cell growth, including the products of tumor suppressor genes and dominant protooncogenes. This protein also contains 3 tandem ankyrin repeats. The BARD1/BRCA1 interaction is disrupted by tumorigenic amino acid substitutions in BRCA1, implying that the formation of a stable complex between these proteins may be an essential aspect of BRCA1 tumor suppression. This protein may be the target of oncogenic mutations in breast or ovarian cancer. Multiple alternatively spliced transcript variants encoding different isoforms have been found for this gene. The threonine at codon 734 is replaced by alanine, an amino acid with similar properties. This alteration has been demonstrated to be defective in the inhibition of mRNA 3' cleavage and degradation of RNAP II in response to DNA damage. Furthermore, UV-induced activation of deadenylation decreased by almost 50% in nuclear extracts from cells expressing p.T734A compared to wild type (Kim HS et al. Cancer Res. 2006 May; 66(9):4561-5; Cevher MA et al. EMBO J. 2010 May; 29(10):1674-87). Structural analysis shows that this residue is buried within a BRCT repeat, suggesting that loss of function in p.T734A may be due to its negative effects on the integrity of the BRCT repeat structure rather than loss of a phosphorylation site (Edwards RA et al. Biochemistry. 2008 Nov; 47(44):11446-56), although direct evidence is unavailable. This amino acid position is highly conserved in available vertebrate species.
