PALB2 c.2480_2481delCA has been observed in at least one individual with breast cancer (Wang 2018). We consider this variant to be pathogenic. -

BMC Cancer, 2018 03;18:315). This alteration as also been reported with a carrier frequency of 0.0001 in a cohort of 12,490 male controls of Japanese ancestry (Momozawa Y et al. Nat Commun, 2018 10;9:4083). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
