Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. ClinVar contains an entry for this variant (Variation ID: 67989). This missense change has been observed in individual(s) with long QT syndrome and Brugada syndrome (PMID: 9686753, 20102920, 26304620). It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces aspartic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 1790 of the SCN5A protein (p.Asp1790Gly). - |
