In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 62 of the KCNH2 protein (p.Arg62Gln). This variant is present in population databases (rs199473664, gnomAD 0.001%). This missense change has been observed in individual(s) with clinical features of KCNH2-related conditions (PMID: 18752142, 29881912). ClinVar contains an entry for this variant (Variation ID: 67298). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Experimental studies have shown that this missense change affects KCNH2 function (PMID: 29881912).
This missense variant replaces arginine with glutamine at codon 62 of the KCNH2 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373).
