Variant summary
Our verdict is Pathogenic. The variant received 12 ACMG points: 12P and 0B. PVS1PM2PP5_Moderate
The NM_004614.5(TK2):c.142dupG(p.Glu48GlyfsTer102) variant causes a frameshift change involving the alteration of a non-conserved nucleotide. The variant was absent in control chromosomes in GnomAD project. Variant has been reported in ClinVar as Pathogenic (★). Synonymous variant affecting the same amino acid position (i.e. E48E) has been classified as Likely benign. Variant results in nonsense mediated mRNA decay.

Mitochondrial disease Pathogenic:1
TK2 p.Glu48GlyfsTer102 (c.142dup) is a frameshift variant that results in the production of a truncated protein which is predicted to have a deleterious effect TK2 gene function. It is also described as c.268dupG, fs191X, fs149X, and E90GfsX102 in the literature. This variant has been observed in a proband affected with mitochondrial disease (PMID:12655576). This variant is not present at a significant frequency in gnomAD. In conclusion, we classify TK2 p.Glu48GlyfsTer102 (c.142dup) as a pathogenic variant. -
