The encoded protein, which is localized to the endoplasmic reticulum, is involved in transferring ethanoloamine phosphate to mannose 2 of glycosylphosphatidylinositol species H7 to form species H8. Allelic variants of this gene have been associated with intellectual disability, hypotonia, and early-onset seizures. Alternative splicing results in multiple transcript variants. In summary, this variant is a rare missense change that is not predicted to affect protein function, and is found in the population at an appreciable frequency. This variant is not anticipated to cause disease; however, the available evidence is currently insufficient to prove that conclusively. Therefore, it has been classified as a Variant of Uncertain Significance. This variant is present in population databases (rs374262167, ExAC 0.05%) but has not been reported in the literature in individuals with a PIGG-related disease. This sequence change replaces alanine with valine at codon 723 of the PIGG protein (p.Ala723Val). The alanine residue is moderately conserved and there is a small physicochemical difference between alanine and valine. -
