Variant summary
Our verdict is Pathogenic. Variant got 12 ACMG points: 12P and 0B. PVS1PM2PP5_Moderate
The NM_001009944.3(PKD1):c.8590G>T(p.Glu2864*) variant causes a stop gained change involving the alteration of a non-conserved nucleotide. The variant was absent in control chromosomes in GnomAD project. In-silico tool predicts a pathogenic outcome for this variant. Variant has been reported in ClinVar as Pathogenic (★). Variant results in nonsense mediated mRNA decay.

The PKD1 c.8590G>T; p.Glu2864Ter variant has been reported in at least 2 individuals affected with autosomal dominant polycystic kidney disease (Liu 2015). It is absent from general population databases (1000 Genomes Project, Exome Variant Server, and Genome Aggregation Database), indicating it is not a common polymorphism. This variant induces an early termination codon and is predicted to result in a truncated protein or mRNA subject to nonsense-mediated decay. Based on the above information, this variant is considered pathogenic. References: Liu Y et al. Targeted Next-Generation Sequencing for Clinical Diagnosis of 561 Mendelian Diseases.
