Variant summary: CFTR c.489G>A (p.Lys163Lys) alters a conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Several computational tools predict a significant impact on normal splicing: Three predict the variant abolishes a 5' splicing donor site. One predict the variant weakens a 5' donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 245706 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.489G>A has been reported in the literature in individuals affected with Cystic Fibrosis (Macek_1997). These data do not allow any conclusion about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 9150159).
