The p.S1164I variant (also known as c.3491G>T), located in coding exon 9 of the BRCA1 gene, results from a G to T substitution at nucleotide position 3491. The serine at codon 1164 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

ClinVar contains an entry for this variant (Variation ID: 54900). This missense change has been observed in individual(s) with breast cancer and ovarian cancer (PMID: 18680205, 25896959). This variant is not present in population databases (ExAC no frequency). This sequence change replaces serine with isoleucine at codon 1164 of the BRCA1 protein (p.Ser1164Ile). The serine residue is highly conserved and there is a large physicochemical difference between serine and isoleucine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Experimental studies are conflicting or provide insufficient evidence to determine the effect of this variant on BRCA1 function (PMID: 18680205, 19493677).
