When cloned, it was discovered to be a human homolog of the E. coli mismatch repair gene mutS, consistent with the characteristic alterations in microsatellite sequences (RER+ phenotype) found in HNPCC. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251440 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1935A>C has been reported in the literature in an individual from a cohort of patients primarily affected with Lynch Syndrome-associated cancers (Li_2020). This report does not provide unequivocal conclusions about association of the variant with Lynch Syndrome. A functional study which used deep mutational sequencing combined with an in vitro DNA mismatch repair (MMR) assay to examine MSH2 missense variants showed no damaging effect of this variant on MMR activity (Jia_2021). The following publications have been ascertained in the context of this evaluation (PMID: 31391288, 33357406). Three clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 and classified the variant as either VUS (n=2) or likely benign (n=1).
