The W1954X nonsense variant in the NSD1 gene has been reported previously to have occurred de novo in at least one individual with Sotos syndrome (Douglas et al., 2003). This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The S1954X variant is not observed in large population cohorts (Lek et al., 2016). Based on currently available evidence, we consider W1954X to be pathogenic. -
