Breast-ovarian cancer, familial, susceptibility to, 2 Pathogenic:2
Variant allele predicted to encode a truncated non-functional protein. -
This sequence change creates a premature translational stop signal (p.Lys1530Glufs*4) in the BRCA2 gene. It is expected to result in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency). ClinVar contains 5 entries for this variant (Variation ID: 216856) all of which interpret this variant as pathogenic. Loss-of-function variants in BRCA2 are known to be pathogenic (PMID: 20104584). Therefore, this variant has been classified as Pathogenic. -
not provided Pathogenic:2
Frameshift variant predicted to result in protein truncation or nonsense mediated decay in a gene for which loss of function is a known mechanism of disease; Truncating variants in this gene are considered pathogenic by a well-established clinical consortium and/or database; Identified in an individual with family history of pancreatic cancer (Murali et al., 2021); Not observed at significant frequency in large population cohorts (gnomAD); Also known as 4815dupG; This variant is associated with the following publications: (PMID: 20104584, 26295337, 34399810) -
The BRCA2 c.4587dup (p.Lys1530Glufs*4) variant alters the translational reading frame of the BRCA2 mRNA and causes the premature termination of BRCA2 protein synthesis. This variant has been reported in the published literature in an individual with a family history of pancreatic cancer (PMID: 34399810 (2021)), and observed in an individual and family member with breast cancer in our internal patient population. This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Based on the available information, this variant is classified as pathogenic. -
Hereditary breast ovarian cancer syndrome Pathogenic:2
Variant summary: The BRCA2 c.4587dupG (p.Lys1530Glufs) variant results in a premature termination codon, predicted to cause a truncated or absent BRCA2 protein due to nonsense mediated decay, which are commonly known mechanisms for disease. Truncations downstream of this position have been classified as pathogenic by our laboratory (e.g.c.4965C>G/p.Tyr1655X, c.5073dupA/p.Trp1692fs). One in silico tool predicts a damaging outcome for this variant. This variant is absent in 120446 control chromosomes from ExAC. In addition, multiple clinical diagnostic laboratories/reputable databases classified this variant as pathogenic. Taken together, this variant is classified as pathogenic. -
This sequence change creates a premature translational stop signal (p.Lys1530Glufs*4) in the BRCA2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in BRCA2 are known to be pathogenic (PMID: 20104584). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with BRCA2-related conditions. ClinVar contains an entry for this variant (Variation ID: 216856). For these reasons, this variant has been classified as Pathogenic. -
Hereditary cancer-predisposing syndrome Pathogenic:1
The c.4587dupG pathogenic mutation, located in coding exon 10 of the BRCA2 gene, results from a duplication of G at nucleotide position 4587, causing a translational frameshift with a predicted alternate stop codon (p.K1530Efs*4). This alteration was identified in a patient with a family history of pancreatic ductal adenocarcinoma (Murali K et al. Hered Cancer Clin Pract, 2021 Aug;19:33). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
