The c.3113A>G (p.Y1038C) alteration is located in exon 4 (coding exon 4) of the MSH6 gene. This alteration results from a A to G substitution at nucleotide position 3113, causing the tyrosine (Y) at amino acid position 1038 to be replaced by a cysteine (C). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
This missense variant replaces tyrosine with cysteine at codon 1038 of the MSH6 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual with suspected Lynch syndrome (PMID: 22495361). This variant has also been identified in 2/248990 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
