The encoded protein is a member of the MRE11/RAD50 double-strand break repair complex which consists of 5 proteins. This gene product is thought to be involved in DNA double-strand break repair and DNA damage-induced checkpoint activation. [provided by RefSeq, Jul 2008]

This variant has not, to our knowledge, been published in the literature as pathogenic or benign. NBN Ile108Thr was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Since Isoleucine and Threonine differ in polarity, charge, size or other properties, this is considered a non-conservative amino acid substitution. NBN Ile108Thr occurs at a position where amino acids with properties similar to Isoleucine are tolerated across species and is located in the BRCT domain (Uniprot). In silico analyses are inconsistent regarding the effect this variant may have on protein structure and function. Based on currently available information, it is unclear whether NBN Ile108Thr is pathogenic or benign. We consider it to be a variant of uncertain significance. -
