This variant is also reported in ClinVar (Variation ID: 469394) and is found in the general population with an overall allele frequency of 0.002% (6/270786 alleles) in the Genome Aggregation Database. Computational analyses predict that this variant is neutral (REVEL: 0.142). However, this variant is located in the exonuclease domain (Palles 2013), and gene-disease association has been established for variants within the exonuclease domain (Seifert 2019). Due to these contrasting evidences and the lack of functional studies, the clinical significance of the p.Arg306His change remains unknown at this time. - |
