Thus, it is implicated in the control of both DNA replication and transcription. Deletion of this gene has been associated with myelodysplastic syndrome and acute myelogenous leukemia. The c.745delG mutation causes a premature Stop codon at position 249, denoted p.V249X. This mutation is predicted to cause loss of normal protein function through protein truncation. The c.745delG mutation was not observed in approximately 6500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. We interpret c.745delG as a disease-causing mutation. -
