The BRCA2 c.2860_2862del p.(Glu954del) change deletes three nucleotides at position 2860-2862 resulting in an in-frame deletion of one amino acid at codon 954. This variant has a maximum subpopulation frequency of 0.0009% in gnomAD v2.1.1 (https://gnomad.broadinstitute.org). This variant was reported in a high-risk breast and/or ovarian cancer family who also had a pathogenic BRCA1 variant (PMID: 16905680). In summary, the evidence currently available is insufficient to determine the clinical significance of this variant. It has therefore been classified as of uncertain significance. -

The c.2860_2862delGAG variant (also known as p.E954del) is located in coding exon 10 of the BRCA2 gene. This variant results from an in-frame GAG deletion at nucleotide positions 2860 to 2862. This results in the in-frame deletion of a glutamic acid at codon 954. This variant has been reported in one high risk breast/ovarian cancer family; however, this family also carried a pathogenic mutation in BRCA1 (Simard J et al. J. Med. Genet., 2007 Feb;44:107-21). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
